WHEN TO USE
Bio DB AGAR is a culture plate suitable for the control of the processes of production and maturation of fermented substances. Its particular composition allows for the selective growth of yeasts of the genera Brettanomyces and Dekkera. Its specific composition is the result of a comparative study of the selective development of Dekkera and Brettanomyces compared with the microflora present in the cellar. The reliability of the DB AGAR plate is checked with molecular biology techniques (RAPD-PCR, PCR specific species).
DB AGAR is ready for use. It does not require any initial preparation, such as rehydration in the case of freeze-dried plates, or the pouring on absorptive pads of liquid media packaged in single doses. Therefore, no manipulations are necessary that could cause contamination. Because it is in the form of agar, DB AGAR guarantees a better result thanks to the greater amount of medium present in each dish (about 15 mL per dish). The enological sector is particularly interested in the isolation and identification of yeasts belonging to the genera Dekkera and Brettanomyces, because they are responsible for the appearance of off-flavors such as ‘stable’, ‘leather’, ‘horse sweat', etc.
HOW TO USE
BRETT AGAR is a ready-to-use substrate. It can be used in all cases in which detection of a possible presence of micro-organisms is required.
When the concentration of cells per sample is high, and, therefore, a small amount of sample is sufficient for analysis, the sample can be spooned directly onto the surface of the plate. When the number of cells is very low, a preliminary concentration of the sample must be put on a microporous membrane, and then incubated on the surface of DB AGAR (as in checking microbiological stability in a bottled sample). DB AGAR can also be used to check surfaces through collecting with a sterile swab and then smearing on the agar.
For the growth of the yeasts, the use of black membrane filters and incubation times of at least 5-8 days at a temperature of 30 °C are recommended. The presence of the yeasts is at first noticed as the color of the plate changes from green to yellow, and then the white-colored colony appears.
